Peptide conjugation to the carrier protein

Instead of the commonly used carrier protein KLH, we use a hemocyanin from a different species: Concholepas concholepas. This carrier protein was developed by Biosonda in Chile and is known as Blue Carrier Protein (BCP). Nowadays, we can find many vendors selling this carrier as it is excellent for generating peptide antibodies.

Most of our peptides are conjugated to this carrier protein by the chemical linker EDC. This is carried out in MES buffer at pH4.5. However, some peptides will not be fit for conjugation at this pH and then we will use linker MBS in phosphate buffer at pH7.

After the cross-linking the mixture is thoroughly dialyzed against PBS in dialysis tubing with MW cut-off value of 12000.
