EB11681 1
EB11681 Immunofluorescence analysis of paraformaldehyde fixed Jurkat cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10ug/ml) followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing plasma membrane and nuclear staining. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10ug/ml) followed by Alexa Fluor 488 secondary antibody (2ug/ml).
EB11681 2
EB11681 Immunofluorescence analysis of paraformaldehyde fixed U2OS cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10ug/ml) followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing plasma membrane, nuclear and cytoplasmic staining. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10ug/ml) followed by Alexa Fluor 488 secondary antibody (2ug/ml).

Target Protein

Principal Names: CD34, CD34 antigen, CD34 molecule, CD34-T, hematopoietic progenitor cell antigen CD34, OTTHUMP00000034733, OTTHUMP00000034734
Official Symbol: CD34
Accession Number(s): NP_001764.1
Human GeneID(s): 947
Non-Human GeneID(s): 12490 (mouse)
Important Comments: This antibody is expected to recognize reported isoform CD34-T (NP_001020280.1) only.

Immunogen

Peptide with sequence C-SPTGERLELEP, from the C-Terminus of the protein sequence according to NP_001764.1.

Please note the peptide is available for sale.

Purification and Storage

Purified from goat serum by ammonium sulphate precipitation followed by antigen affinity chromatography using the immunizing peptide.
Supplied at 0.5 mg/ml in Tris saline, 0.02% sodium azide, pH7.3 with 0.5% bovine serum albumin.
Aliquot and store at -20°C. Minimize freezing and thawing.

Applications Tested

Peptide ELISA: antibody detection limit dilution 1:128000.
Western blot: Preliminary testing showed bands at approx 85+90kDa in Human Peripheral Blood Mononucleocytes lysate (calculated MW of 35.0kDa according to NP_001764.1). The observed MW correspond to glycosylated forms, routinely observed by other sources. Primary incubation 1 hour at room temperature.

Immunofluorescence: Strong expression of the protein seen in the plasma membranes and nuclei of Jurkat and U2OS cells. Recommended concentration: 10µg/ml.

Flow Cytometry: A customer reported weak staining of Jurkat cells at a concentration of 10ug/ml.

Species Reactivity

Tested: Human
Expected from sequence similarity: Human, Mouse
