Purification and Storage

Purified from goat serum by ammonium sulphate precipitation followed by antigen affinity chromatography using the immunizing peptide.
Supplied at 0.5 mg/ml in Tris saline, 0.02% sodium azide, pH7.3 with 0.5% bovine serum albumin.
Aliquot and store at -20°C. Minimize freezing and thawing.

Applications Tested

Peptide ELISA: antibody detection limit dilution 1:16000.
Western blot: Approx 30kDa band observed in Human and Mouse Thymus and Human Spleen lysates and both a 30kDa and 35kDa band observed in Human Lymph node lysates, both blocked by incubaion with the immunising peptide (calculated MW of 28.3kDa according to Human NP_001166039.2, 34.1kDa according to Human NP_001166038.2 and 33.8kDa according to Mouse NP_034981.1). Recommended concentration:0.1-0.3µg/ml. Primary incubation 1 hour at room temperature.
IHC: In paraffin embedded Human Tonsil shows staining of clusters of cells some of which are likely plasma cells. Recommended concentration: 4-6µg/ml.

Immunofluorescence: Strong expression of the protein seen in the nuclei and cytoplasm of U2OS and Jurkat cells. Recommended concentration: 10µg/ml.

Flow Cytometry: Flow cytometric analysis of Jurkat cells. Recommended concentration: 10ug/ml.
